Abstract
This study investigated the promoter elements (Pfam) of the target genes of tetracycline-inducible expression plasmid inE. coli. The sequence of pTA4-tetA-1 was chosen as a novel promoter sequence and a plasmid containing a tetracycline-responsive promoter (TRE) was constructed. All the target genes were under the control of the tetO-GAL4-tetA promoter in the presence of tetracycline (TC) in the media of an exponential growth phase. The presence of tetracycline-inducible promoters was confirmed through PCR analysis with the tetracycline-inducible promoter sequence and a tetracycline-responsive promoter sequence in apET14 strain. The tetO-inducible promoter element was expressed in the presence of tetracycline-inducible promoters in the media ofpET14 and tetO-TRE-TRE. TetO-inducible promoters were constructed in the presence of tetracycline-inducers in the media of an exponential growth phase. The tetO-inducers were used as a control to control the expression of Tet-1 in the absence of tetracycline.
